Mindmaxxing — Semax, Selank, DSIP & CNS Neuropeptides

Mindmaxxing collects the peptides small enough and stable enough to reach the central nervous system, which is a far shorter list than the nootropics market implies. Most of it came out of Soviet and Russian research programmes from the 1970s onward and carries trial data that never crossed into Western literature in any organised way. Semax is ACTH(4-10)-derived and studied for BDNF upregulation and sustained attention, while Selank comes from tuftsin and is studied for anxiolysis through GABAergic modulation without sedation or tolerance. DSIP addresses sleep architecture rather than sedation, and PE-22-28 is the newest thing here, a TREK-1 channel inhibitor with rodent depression-model data and nothing human behind it yet. Why the N-Acetyl Amidated Forms Exist

Native Semax and Selank are both short peptides with serum half-lives measured in minutes, which is a problem for anything that has to survive long enough to cross the blood-brain barrier. Acetylating the N-terminus and amidating the C-terminus blocks the exopeptidases that chew a peptide from both ends, and the result is a meaningfully longer window of activity from the same dose. That is why N-Acetyl Semax Amidate and NA-Selank Amidate are stocked alongside the plain versions rather than instead of them. The tradeoff is cost per vial, and for intranasal administration — a short path with less enzymatic exposure — the plain forms remain defensible.

DSIP Is a Sleep Architecture Compound

Delta Sleep-Inducing Peptide is named for what it was isolated in connection with, not for a sedative effect it does not have. The interest is in slow-wave sleep proportion — the delta-frequency stage where growth hormone release concentrates and where most of the restorative work of a night happens. It does not knock anyone out, and people expecting a hypnotic are usually disappointed. Semax and the BDNF Question

Semax is a synthetic fragment of ACTH(4-10) with a Pro-Gly-Pro tail added for stability, and the mechanism most often cited for it is BDNF and NGF upregulation in the hippocampus. The Russian clinical record covers stroke recovery, cognitive impairment and optic nerve conditions, and Semax carries a registered indication there that it does not have anywhere else. Western replication is thin, which is the honest caveat: the evidence base is real but it is concentrated in one research tradition and one language.

PE-22-28 and the TREK-1 Route

PE-22-28 is a spadin analogue that blocks the TREK-1 potassium channel, and the reason that is interesting is timeline. TREK-1 knockout produces a depression-resistant phenotype in rodents, and pharmacological blockade appears to reproduce antidepressant-like effects over days rather than the weeks SSRIs require. What is the difference between Semax and N-Acetyl Semax Amidate?

N-Acetyl Semax Amidate is the same peptide with the N-terminus acetylated and the C-terminus amidated. Both modifications block exopeptidase degradation, so the compound survives longer in serum and delivers a longer window of activity from an equivalent dose. Plain Semax is cheaper and remains reasonable for intranasal use, where the path is short and enzymatic exposure is lower.

Does DSIP work as a sleep aid?

Not in the way a hypnotic does. DSIP is studied for its effect on sleep architecture — specifically the proportion of slow-wave, delta-frequency sleep — rather than for sedation. It will not put anyone to sleep. The interest is in what happens to the quality of sleep already occurring, which is also where growth hormone release concentrates.
